Could I use a 5v to 12v step up module connected to 1000c to power a 12v screen

Which screen? It's very likely your 12v screen has a step-down module to bring the voltage to 5v, as the panel will run off 5v.

Modifying the board to bypass the step-down module is what's needed to run the screen off 5v.

[edit] Never mind, I just noticed your previous posts. I'm guessing you mean a different screen since you're already aware of the modifications required.

I would think having two step up converters in a series will introduce even more distortion that perhaps may give the screen weird artifacts.

Yes, you can. That is what i am doing with my 640x480 VGA Display.
The Pololu U1V10F5 works fine and is really small.
